Femtosecond Dynamics of Rhodopsin

Abstract

In the present study we clarify the detailed mechamism of the excited state dynamics of rhodopsin followed by the formation of the primary photoproduct by femtosecond spectroscopy in 400-1000 nm probe spectral region using tunable femtosecond laser over 500-600 nm obtained by using two-stage dye amplifiers of wavelength-selected femtosecond continuum. The experiment has improved the data dramatically in time resolution in comparison with our previous work1 and in the probe spectral region in comparison with the very recent work at U. C. Berkley2. The output pulse width and energy are 300 fs, > 2µJ for spectral width of 3 nm. Octopus rhodopsin solubilized by detergent(L-1690 or SM-1200) in H2O or D2O was used as samples.
