Developing 3D high-resolution imaging of pancreatic islet β cells in genetically obese-diabetic (ob/ob) mice

Abstract

We characterize Fp, NADH and Pyro-2DG fluorescence intensity distributions of pancreatic islet β cells and liver cells in obese-diabetic (ob/ob) mice. A cryo-imager is used to image the pancreas and liver at the following Ex/Em wavelength pairs: 440/525 nm, 365/460 nm and 405/670 nm. We examine the redox characteristic of the pancreatic islet β cells and observe the accumulation of Pyro-2DG in pancreatic islet β cells, then compare both of these with those in liver cells. Construct high-resolution 3D images basing on the 2D images of the serial sections of pancreas. This study helps us evaluate the validity of optical studies of pancreatic tissue.
