Non-invasive single cell tomography on skin cells

Abstract

Non-invasive single cell tomography offers 3D view of cells for quantifiable information, such as nuclear-cytoplasmic ratio and organelle distribution in the cell. It can facilitate the observation of cell proliferation, cell to cell interaction, etc. Cell-to-cell variability might be attributed to genetic, epigenetic, proteomic factors, presence of cancer stem cells at different stages of cell cycle. Measuring response at single-cell level provides pharmacokinetic and pharmacodynamics information to aid drug development and regimen design. In this paper, we report the use of parametric ultrahigh-resolution optical coherence tomography (UR-OCT) to distinguish keratinocytes (HaCaT) and fibroblasts (Hs68) at cellular level, since keratinocyte and fibroblast are the major cells in epidermis and dermis. Total of 64 HaCaT and Hs68 cells were clearly examined by a crystalline fiber based UR-OCT system [1]-[2]. Characteristic parameters of these cells were extracted. Linear discriminant analysis was used to classify HaCaT and Hs68.
