Review of time-Domain Diffuse Correlation Spectroscopy: from theory to human subject studies

Abstract

Time-Domain Diffuse Correlation Spectroscopy (TD-DCS) is a novel approach that aims at merging Diffuse Correlation Spectroscopy (DCS) with time-domain near-infrared spectroscopy (TD-NIRS) in a single modality [1]. The idea is to excite the tissue with a long-coherence, pulsed laser and acquire the temporal point spread function (TPSF) through the measurement of the photons time-of-flight (ToF). The optical properties of the tissue are recovered by TD-NIRS analysis, while DCS processes the temporal speckle fluctuations due to the moving scatterers in tissue (blood cells) to measure an index of microvascular blood flow (BFi) [2]. Furthermore, the time-domain information allows us discriminate between short and long photon paths to perform NIRS and DCS processing only on photons that have travelled deep into the tissue, increasing the sensitivity to the brain. This discrimination allows us to shorten the source-detector separation, increasing the signal-to-noise ratio, and making the optical sensor more compact.
