Underpinning the microvascular origin of BOLD-fMRI with two-photon microscopy

Abstract

We have developed a model of BOLD-fMRI from first principles using from two-photon microscopic measurements of cerebrovascular physiology. The model was validated against experimental BOLD measurements at several levels. Our bottom up approach predicted a new phenomenon: the folding of the cortical surface produces variations in BOLD signal corresponding to 40% of the amplitude of the BOLD signal generated by neuronal activation. This prediction was further validated experimentally with human BOLD-fMRI measurements, confirming the predictive power of our approach.
