Spectral Decomposition of Multicolor Imaging in Multifocal Multiphoton Microscopy

Abstract

The study of synapse formation and dendrite remodeling is critical in understanding the mechanisms underlying brain plasticity. Spectral resolved multiphoton microscopy is required to map synapse and dendrite structures in the brain of living mice. For imaging of multiple fluorophores, spectral decomposition is often necessary due to the spectral overlap of emission signals. While linear and nonlinear decomposition techniques have been developed, further improvement in spectral separation may be possible by the inclusion of Poisson noise removal.
